peptide frequently asked questions:
q: Can the same antibody be used for immunohistochemistry and Western Blot?
Answer: In immunohistochemistry, antibodies recognize antigenic determinants (also called epitopes) that have not been denatured. Some epitopes are linear while others are conformational. Linear epitopes are not affected by protein denaturation. Both natural proteins and boiled proteins contain conformational epitopes. Due to the limitation of protein space structure, denaturation after cooking will disappear. If the antibody you use recognizes several consecutive amino acids on the protein, that is, linear epitopes, then this antibody can be used for immunohistochemistry and Western at the same time, while if the antibody recognizes conformational epitopes, it can only be used for immunohistochemistry.
