Background and Overview
Etelcalcetide,another name is called AMG416.In previous reports, Fmoc-L-Cys(Npys)-OH, Boc-L-Cys(Npys)-OH, H-L-Cys(Npys)-OH, and other special protective amino acids were used. Due to their high cost, forming disulfide bonds on solid-phase resin has many unfavorable factors such as excess and difficulty in control. Therefore, low-cost H-L-Cys-OH is used as the synthetic material for the final formation of intermolecular disulfide bonds. Several simple oxidation methods are employed in the liquid phase to obtain a high-purity solution of Velacap. The reaction is completed not only in a homogeneous manner but also greatly reduced and controlled in dosage.
The application of Etelcalcetide
Etelcalcetide is a novel calcimimetic agent that has been developed for chronic kidney disease(CKD) patients receiving hemodialysis,administered intravenously. AMG416 binds to and activates the calcium-sensing receptors on the parathyroid glands, leading to a reduction in parathyroid hormone (PTH) levels. It holds great promise as a new drug for effectively treating secondary hyperparathyroidism. The FDA has approved the novel calcimimetic veracelipt for the treatment of secondary hyperparathyroidism in adult hemodialysis patients, marking the first approval of a drug for this condition in dialysis patients in nearly 12 years. Patients with moderate to severe secondary hyperparathyroidism undergoing hemodialysis showed a significant reduction in PTH levels after treatment with veracelipt.The efficacy of veracelipt is superior to placebo and is not lower than cinacalcet.
