Growth Hormone-Releasing Peptide-6 (GHRP-6) is a synthetic peptide that promotes the secretion of growth hormone. It stimulates the release of endogenous growth hormone (GH) by specifically binding to receptors in the pituitary gland and hypothalamus, thereby exerting various physiological effects.
Side effects
Growth hormone releasing peptide-6 has mild side effects on the human body, such as flushed face, slight sweating, and mild drowsiness. Some children may occasionally experience changes in serum tests, adrenal cortical hormones, and prolactin.
Preparation
Peptide synthesis is performed under standard conditions using Rink resin in an automatic shaker. HBTU (3 equivalents) is used as the coupling reagent, and DIEA (initially 3 equivalents, then 1 equivalent 20 minutes after the start of the coupling reaction) is used as the base to carry out the amino acid coupling (3 equivalents) in DMF. Fmoc deprotection is done by treating the resin with 20% piperidine in DMF for 10 and 20 minutes. After each coupling and deprotection step, the resin is washed with DMF (2x), MeOH (2x), and DCM (2x) respectively. Side chain deprotection and Aza-Peptide cleavage are performed. The resin is then treated with a freshly prepared TFA/H2O/TES solution (95:2.5:2.5, v/v/v, 20mL/g Aza-peptide resin) at room temperature for 2 hours. The cleavage mixture is filtered, and the resin is washed with neat TFA. The filtrate is concentrated to about 1 ml and treated with Et2O. The resulting N-heterocyclic peptide precipitate is filtered, washed with Et2O, dissolved in an acetonitrile/water (1:1, v/v) solution, and lyophilized to yield a foam or powder of Growth Hormone-Releasing Peptide-6.
