The Function of the P21 Peptide

    P21 Peptide (P21 peptide) is an artificially synthesized CNTF (ciliary neurotrophic factor) analog that can promote neuron growth and shows significant potential in drug development, cell cycle regulation, tumor inhibition, neuroprotection, and aging-related research. The basic sequence of P21 peptide is Ac-DGGL-NH2. To enhance the peptide's stability and its ability to cross the blood-brain barrier, researchers introduced an adamantane structural unit at the C-terminus of the peptide. As a result, the derived P21 peptide may be able to mimic the effects of CNTF by activating the CNTF receptor complex and its downstream signaling pathways.

    P21 peptide can promote neuron survival, differentiation, and synapse formation by activating neurotrophic factor signaling pathways. The P21 peptide is closely associated with the tumor suppressor protein p53 and, as a downstream gene of p53, mediates G1 phase cell cycle arrest through a p53-dependent pathway in response to stress signals such as DNA damage.

Tumor Suppression Potential:

 As a CDK inhibitor, the P21 peptide can block the tumor cell cycle progression and induce apoptosis by inhibiting its activity. The P21 peptide can be used in combination with chemotherapy drugs to enhance tumor treatment. At the same time, p53-P21 targeted drugs are under development.
